Both the previous generation 500x and new generation 500x2 (as well as other models) support ISO and BD-ISO. However, it should be noted that the menu support for BD-ISO is not comprehensive. My complete DVD collection is loaded as ISOs on my NAS and the 500x as well as 500x2 has never had an issue playing any of them.

Speaking of a NAS, can anybody tell me why I cannot use the menu while playing movies from my NAS [Synology] to my Mede8er?
The movie plays well if it has no menu.

What kind of movies do you mean exactly?
Blue-Ray or DVDs with a menu?

A 500X has no Blue-Ray menu support as far as I am aware.
An X2 and X3 have, but only BD-light, which means it cannot play Blue-Ray menus with Java, only menus without.

If you are talking about DVD menus, that is possible with all, but will only work when streaming from a NAS when you use filesharing (samba) streaming or NFS streaming. Upnp streaming does not support menus.
